On the fifth day, Sasuke was harnessing the chakra of Kurama while meditating to manifest Senjutsu chakra.
Difference for today however, he was using in absence of his normal chakra.
Sasuke took Kurama chakra from Naruto, who had yang side of the original.
Considering you need yang chakra to use medical ninjutsu, in theory if he use Kurama (yang) chakra without being diluted by his chakra, supposedly not sufficient at converting into yang chakra, he might get a shot at succeeding with Senjutsu chakra to strengthen and speed up the process.
Of course, he was doing it around 2am as explaining to Kakashi how he got Kurama going to be a hassle, and might even ruin image to others.
'Though calling mystical palm technique A rank jutsu is quite misleading.' Sasuke sighed at Tsunade's supposed pettiness, learning he would win the bet if he was able to use it for straight 5 seconds.

In simplest terms, it will increase regeneration of cells in the place the wound was inflicted.
It same applies to undesirable cells that were present however, they would worsen patients' condition.
It includes poison, allergies, cancer cells etc.
And it doesn't do anything against diseases a patient has.
So medical-nin had to learn tons of knowledge so that able to recognise and understand the disease and treat it with ingredients available to you and in the environment and shops, and treat them similar to medical doctors in ancient times such as China and India or in advance.
In times where mythical plan technique is needed to heal them such as during surgery, extreme precision is crucial, where they have to be careful to use it on correct cells over harmful cells.
This is precisely why Tsunade created 3 rules medical-nins must obey, as their combat capability would decline considerably while honing medical knowledge.
At last, duration to maintain is important as using to heal for 30 minutes is better than using the jutsu multiple times to get same duration, hence duration is indicator of
proficiency of use.
While thinking about medical ninjutsu, his thoughts unexpectedly drifted to the root.
'If Danzo wasn't that extreme in ideas and actually thought of making a powerful and well functioning hidden organisation, he would treasure Nono Yakushi and Kabuto Yakushi similarity to Fū and Torune.'

Why cause massacre and decrease the manpower of the village rather than indoctrinating them to Shinsui level?
Sasuke understood why he and alongside others 3 wanted Elite Uchiha to be dead as they won't listen to the elder council but why he even included Uchiha civilians children and children of the elite Uchuha who would be easier to indoctrinate and imprint blind loyalty for the 'village'?
If they are able to imprint their 'will of fire' on Shisui and Itachi, why not on others?
Or his and their hate towards Uchiha was so great that they preferred a ethnic cleanse?
He was curious whether Kumogakure and others did attack Konoha at one point, after hearing of the death of one of their strongest clans.
He was also curious why didn't Obito didn't attack Konoha when he was controlling the 4th Mizukage.

While Sasuke was trying his best to perform the Mystical Palm Technique in his room, Tsunade observed him from a distance, outside the inn in a secluded part of the forest.
'Why does he feel familiar?' she thought, feeling as if a fragment of her grandparents lingered—sensing the essence of Sasuke's use of Senjutsu and Kurama's chakra, energies each of her grandparents once wielded
However, she was not a sensor-type, so she couldn't unravel the mystery of Sasuke, a member of the clan her grand-uncle despised most.
Initially, she suspected Sasuke was Fugaku's illegitimate heir—one Itachi hadn't known about. His lack of typical Uchiha traits, coldness,reservation and arrogance, might stem from growing up outside the clan compound.
Otherwise, why would Itachi spare him after slaughtering the rest?
'I suppose returning to the village will shed light on the massacre,' she mused, though she dreaded going back. The village she once called home now lay in decline, mismanaged by her grand-uncle's disciples.
"He's likely regretting his choices in the afterlife," she muttered, imagining his remorse for trusting the wrong people.
Despite her lineage and power to overthrow them, Tsunade had no interest in becoming Hokage. She was a healer, not a politician tasked with judging others' competence.
She respected the role but believed it should go to someone capable, not self-serving elders. Worse, she feared becoming like them—people who'd prioritized their own interests since the Second World War.
Also, she feared to become the very same old folks she dreads, admitting they were once seem fine people until around the second world war, where they began to prioritise their own interest.
"So Lady Tsunade, may I spare some words?"
"Lady Tsunade, may I have a word?" Kakashi appeared behind her, Raijin no Ken resting on his left arm.
Tsunade instinctively about to question Kakashi possession of her grand uncle's blade, but dismissed it, thinking her sensei sent it to appease her like always.
"Speak, I don't have all day." Tsunade said, shrugging off the spook she felt earlier.
"Do you know why Uziogakure died ?" Kakashi's voice darkened as purple lightning erupted from the sword, narrowly missing her.
"Are you trying to kill us?!" she shouted as the blade's glow engulfed them.
"The sword's memories will explain better than I can," Kakashi murmured, activating its hidden power.
